I hate to potentially necropost this thread, but I popped in to see if there has been any progress made on figuring out the runes? Since the OP said runes could be edited the same way as gems, I figured this would be the appropriate place to ask.
I've figured out Moon and Communion, but that's about it—I'm sure I found another one but I might've messed up testing it and wound up with a debug item in my inventory. Guidance has already been posted, so there's no need to post that. Has anyone found any other runes? From what I know, the max tier Moon rune is E2 C8 10 00 while Communion +5 is B4 4D 11 00.

Well, Runes are coded in the same fashion as Gems.
Since you can easily get almost all the runes in a legit way, I decided to give specific instructions only for Guidance 3.
I expect people would grab a 2nd Guidance 1 from NG+1 (this rune spawns each cycle no matter if you already have it or not) and mod it into a Guidance 3 :
Tier 1 : 85 01 80 C0 / 40 96 01 80 / 02 00 00 00 / 01 00 00 00 / 08 69 11 00
Tier 2 : 8D 01 80 C0 / 41 96 01 80 / 02 00 00 00 / 01 00 00 00 / 09 69 11 00
Tier 3 : XX XX XX XX / XX 96 01 80 / 02 00 00 00 / 01 00 00 00 / 0A 69 11 00
My personnal choice is to input Tier 3 this way : XX XX XX XX / 42 96 01 80 / 02 00 00 00 / 01 00 00 00 / 0A 69 11 00
in order to follow the trend of the first two runes. If you choose to mod the "source" part like me, don't forget that it's written twice within the savedata so mod it both times.

Awesome! Got it work. Just a quick question, How safe is it to use it online? Just for co op with a friend who needs help with bosses. Feel kinda weird having a weapon with 9x +33% blood gems lol

I can not simplify any more than I already have.
Advanced Mode requires some basic Hex editing knowledge.

do i have to put my gems in inventory? some of them is in the storage or weapons

The latter doesn't matter, what you want to find is the former. Find the gem, and modify its attributes.

Ok, i guess im doing it wrong cause i have some gem that i cant find, except for 27% physical and boss watcher
